Functional characterization of the putative tumor suppressor genes EMP3 and ST13 in glioma

The major research goal of the subproject 13 is the functional characterization of the putative glioma tumor suppressor genes (TSG) EMP3 on 19q13.3 and ST13 on 22q13 in human cell lines and in a knock-out (KO) mouse model (EMP3). Genes potentially involved in EMP3- and ST13-pathways will be determined by expression profiling and functionally validated. Expression of EMP3, ST13 and potentially dependent genes will be evaluated by IHC in human tissue microarrays (TMA) consisting of tumors of the glioma core collection as well as in frozen human glioma. Expression will be correlated with clinical parameters to test clinical application. Thereby, we will pursue on the NGFN and NGFN2-Brain Tumor Network by functionally characterizing some of the most promising putative TSG on 19q and 22q. The functional characterization of EMP3 and ST13 will help to understand the mechanisms that induce and propagate gliomas and may allow the development of diagnostic assays. We will provide materials and clinical information concerning the NGFN glioma core collection and offer histological evaluation of brain tumor models to all members of the IG.
