The major goal of this subproject is the analysis of modulators of the glutamatergic system in
alcohol addiction, with a special emphasis on the role of D.serine, an allosteric modulator of N-Methyl-D-aspartate receptors (NMDA). D-serine is synthesized from L-serine in astrocytes, through the activity of serine-racemase. It is transported out of the cell though Asc-1 und ASCT2 transporters, and degraded by the enzyme D-amino-acid-oxidase (DAO). Our laboratory has already generated a number of genetic mouse models with a deletion or overexpression of these enzymes. Other missing models will be established in the context of this project. All animals will be comprehensively analyzed for altered physiological and behavioural responses to alcohol.
