ABCC6 and arterial calcification

The overall research goal of this sub-project is to elucidate and functionally characterize genetic determinants of cardiovascular calcification. Indeed, the abcc6 gene was positionally cloned as a major cause of dystrophic calcification. However, the precise molecular mechanisms have not yet been unravelled. A more thorough understanding of pathophysiologic pathways in cardiovascular calcification will discern new options for risk prediction and thus disease prevention as well as identify new therapeutic targets.
for this reason we hypothesize that  the introduction of the wilde type allele of Abcc6 in DCC-susceptible congenic mice will rescue calcification phenotype in the recipient mice. These mice will serve as model to understand in depth the functionel role of Abcc6 in the initiation and devlopment of calcification in both human and mice.
