Modifier Mapping in CSS Strains

The goal of this subproject is the identification and chromosomal mapping of modifier genes for intestinal cancer. Modifiers could be genes that affect the tumor phenotype of the mouse, or genes affecting gene expression levels (hyperactivation) or de novo gene methylation (inactivation) in the intestine, in correlation with intestinal tumor multiplicity and grade induced by the APCmin mutation. To this end, tumor-prone APCmin-mice are crossed to chromosome substitution strains (CSS) carrying Mus.m. musculus strain PWD/Ph (donor) derived single chromosomes on the C57BL/6J (B6) genetic background. This subproject is the basis of the work of most other subprojects. The genetic crosses, histopathological examination of APCmin induced tumors and dissection of wild type intestine and adenomas, is performed in this subproject.
