Recombinant cancer cell libraries & drug target recovery

Commonly, comparative functional and molecular analyses of cancer-relevant genes and their mutated variants have been performed in a rather fragmentary manner using different approaches and model systems.
The goal of this subproject is to utilize standardized systems for site-specific recombination in order to establish recombinant cell line libraries for different cancer types. These resources will be used as a common basis for functional and molecular analyses in IG-MUTANOM.
The identical genomic background of such isogenic recombinant cell lines enables a systematic and quantitative determination of the effects of the selected genes and their mutated variants. In tight interaction with the other subprojects, results of functional analyses will be linked to protein-protein interactions and gene expression patterns as well as to clinical data aiming at the identification of key proteins and their mode of action. Selected candidates will be analysed in detail in order to develop innovative and targeted concepts for cancer therapy.
